This product is a CHO-K1 stable cell line that stably expresses human CCR5 receptor together with β-Arrestin. Upon agonist binding to CCR5, β-Arrestin is recruited to the activated receptor, generating luminescent signal via the split luciferase reporter system. Therefore, receptor activity can be easily quantified in live cells in a dose-response manner by measuring luminescence intensity changes. This cell line is suitable for HTS and receptor functional evaluation assays, as well as antagonist and antibody potency testing targeting CCR5.

Schematic diagram of split luciferase assay: hCCL3 binds CCR5 to induce β-arrestin recruitment and luminescence; Leronlimab blocks hCCL3-CCR5 interaction and inhibits signal output.

Assay Principle

Schematic diagram of β-arrestin recruitment assay for GPCR, agonist triggers β-arrestin recruitment and luciferase complementation, luminescence detected by plate reader for dose-response curve.

Upon agonist binding, the GPCR becomes activated and recruits β-arrestin. The β-arrestin-luciferase fusion protein is brought into proximity with the receptor complex. Addition of substrate restores luciferase activity and generates luminescence signals. The luminescence intensity correlates with the level of β-arrestin recruitment, which can be quantified via a plate reader to generate dose-response curves for ligand characterization.

Flow cytometry histogram showing CCR5 expression in CCR5/β-Arrestin CHO-K1 stable cells (blue) versus parental β-Arrestin CHO-K1 control cells (red) stained with anti‑CCR5 antibody.

Figure 1. Recombinant CCR5/β-Arrestin/CHO-K1 stably expressing CCR5

Blue curve: CCR5/β‑Arrestin/CHO-K1 stained with anti‑CCR5 antibody (Mean FL4‑A = 4.05E5). Red curve: β‑Arrestin/CHO-K1 negative control stained with anti‑CCR5 antibody (Mean FL4‑A = 2177). The obvious shift of fluorescence signal demonstrates high expression level of CCR5 receptor on cell membrane.

Dose-response curve showing Leronlimab inhibition of hCCL3-induced β-arrestin recruitment in CCR5 β-Arrestin CHO-K1 cells, IC50 = 0.25 μg/ml.

Figure 2. Dose response of Human CCL3 in CCR5 β-Arrestin CHO-K1 Cell Line (C18)

The IC50 value was determined to be 0.25 μg/mL. Data shows dose-dependent inhibition of β-Arrestin recruitment by Leronlimab.

Dose-response curve of human CCL3 in CCR5 β-Arrestin CHO-K1 cell line (C18), EC50 = 5.99 ng/ml, S/B = 37.1, measured by luminescent β-arrestin recruitment assay.Dose-response curve of human CCL3 in CCR5 β-Arrestin CHO-K1 cell line (C18), EC50 = 5.99 ng/ml, S/B = 37.1, measured by luminescent β-arrestin recruitment assay.

Figure 3. Inhibition of hCCL3-induced β-Arrestin Recruitment in CCR5 β-Arrestin CHO-K1 Cells (C18)

The EC50 = 5.99 ng/ml with S/B ratio =37.1. The signal was measured as relative light units (RLU). The result demonstrates hCCL3 induces β-Arrestin recruitment in a dose-dependent manner.

Cell Resuscitation
1)Rapidly thaw the frozen cells in a 37 °C water bath for approximately 60 seconds. Once thawed (which may take slightly less or more than 60 seconds), immediately transfer the cell suspension from the cryovial into a 15 mL centrifuge tube containing 10 mL of pre-warmed CHO-K1 Human CCR5 β-Arrestin Cell Line complete culture medium.
2)Centrifuge cells at 1000 rpm for 5 min to remove medium, then resuspend cells in 5 mL of pre-warmed complete medium.
3)Transfer the cell suspension into a T25 culture flask and incubate at 37 °C with 5% CO₂.
4)After approximately 24–36 hours, replace the medium or passage the cells to remove non-adherent dead cells.

Subculturing procedure
1)When the cell density reaches the appropriate confluency for passaging, wash the cells with PBS, then add 1 mL trypsin to detach the cells. When more than 80% of the cells detach upon gently tapping the culture flask, add complete culture medium to terminate digestion. Gently pipette to obtain a single-cell suspension, transfer to a 15 mL centrifuge tube, and centrifuge at 1000 rpm for 5 minutes.

2)Discard supernatant after centrifugation. Resuspend cells in fresh medium to a single-cell suspension and transfer to a new culture flask for continued growth.

Cell Freezing
After trypsinization and centrifugation of cells from each T75 flask or 10 cm culture dish, discard the supernatant. Add 2 mL of cryopreservation medium (90% FBS + 10% DMSO), gently resuspend thoroughly, and aliquot into two cryovials. Immediately place the cryovials into a controlled-rate freezing container (e.g., Nalgene 5100-0001), fill with isopropanol to the indicated level, and store at −80 °C. After 24 hours, transfer the cryovials to liquid nitrogen for long-term storage.
