CRTH2 (DP2, GPR44) is a GPCR mainly expressed on immune cells. Prostaglandin D2 binding activates Gαi/o, inhibiting adenylate cyclase and reducing intracellular cAMP. It mediates type 2 inflammatory responses, closely involved in asthma, allergic rhinitis and atopic dermatitis, and serves as a key drug target for inflammatory diseases.
